The future of phenotypic drug discovery

چکیده
Summary Phenotypic drug discovery (PDD) uses biological systems directly for new screening. While PDD has proved effective in the of drugs with novel mechanisms, broader adoption, key challenges need resolution: progression poorly qualified leads and overloaded pipelines due to lack tools process prioritize hits; advancement undesirable mechanisms that fail at more expensive stages discovery. Here I discuss how human-based phenotypic platforms are being applied throughout hit triage prioritization, elimination hits unsuitable supporting clinical strategies through pathway-based decision frameworks. Harnessing data generated these can also fuel a deeper understanding efficacy toxicity mechanisms. As approaches increase use, they will gain power driving better decisions, generating faster turn promoting greater adoption PDD.

Phenotypic screening: the future of antibody discovery.
Most antibody therapeutics have been isolated from high throughput target-based screening. However, as the number of validated targets diminishes and the target space becomes increasingly competitive, alternative strategies, such as phenotypic screening, are gaining momentum. Here, we review successful phenotypic screens, including those used to isolate antibodies against cancer and infectious ...
متن کاملThe future of crystallography in drug discovery.
INTRODUCTION X-ray crystallography plays an important role in structure-based drug design (SBDD), and accurate analysis of crystal structures of target macromolecules and macromolecule-ligand complexes is critical at all stages. However, whereas there has been significant progress in improving methods of structural biology, particularly in X-ray crystallography, corresponding progress in the de...
متن کاملPhenotypic Drug Discovery with High Content Screening
Drug discovery in its early history used to be conducted mainly in a target-agnostic fashion, focusing on qualitative readouts from a living system -cells, tissue, or organisms -and asking the question, ‘Did this entity affect the phenotype?’. With the rise of molecular biology in the early 1980s and an ever more powerful genomics toolkit from the 1990s, there was a marked shift away from pheno...

This manuscript is a review of a number of ethnobotany and ethnopharmacology texts and papers in order to arrive at current understanding and scope of ethnobotany and ethnopharmacology. It includes either a brief definition of both fields and presenting some examples on them. Also it has a glimpse on the geographical coverage of research works throughout the world, important issue of prope...
متن کاملUnconventional natural sources for future drug discovery.
Natural products used as food or drinks, clues from the fascinating science of zoopharmacognosy, terrestrial animals and their excreta, and many other NPs not primarily known for their medical use, seem to have, to a great extent, escaped the attention of natural product research. It is nevertheless highly probable that these sources could contain biologically active compounds, some of which wi...
